Optimization of Protein Quality Assay in Normal, opaque-2, and Quality Protein Maize

The development of quality protein maize (QPM) was considered a significant leap toward improvement in the nutritional status rural masses developing countries. QPM is attributed to higher concentration essential amino acids, particularly lysine and tryptophan, its kernel endosperm. However, similarity grains normal necessitates standard protocol assess maize. present study aimed at improving assessment QPM. For this purpose, endosperm defatting estimation procedures were restandardized optimized with respect duration amenability for high-throughput analysis. Unlike maize, opaque-2 mutants completely defatted within 48 h period. It observed that tryptophan content, calculated each interval, increased samples longer duration. No differences content analyzed 72 h. Moreover, estimated by using Bradford method certain modifications strongly correlated micro-Kjeldahl ( r = 0.9).
